Presence of Foxp3 Expression in Tumor Cells Predicts Better Survival in HER2-Overexpressing Breast Cancer Patients treated wit

چکیده
Purpose: The Forkhead Box Protein 3 is highly expressed in regulatory T cells, but also in tumor cells, acting as a transcriptional repressor of breast oncogenes including HER2. We investigated the prognostic significance of Foxp3 expression in cancer cells in a large cohort of patients with HER2-overexpressing breast carcinoma treated with neoadjuvant chemotherapy. Methods: Foxp3-positive tumor cells were detected by immunohistochemistry in 103 patients with primary invasive HER2-overexpressing breast carcinoma, and treated with neoadjuvant chemotherapy, with or without trastuzumab. Kaplan-Meier analysis and Cox regression model were used to assess relapse-free and overall survival, according to the presence or absence of Foxp3 expression in tumor cells. Results: Breast cancer cells were Foxp3+ in 57% of tumors. Foxp3 expression in breast cancer cells was associated with better relapse-free (P = 0.005), and overall survival (P= 0.03). By multivariate analysis, presence of Foxp3+ tumor cells resulted an independent prognostic factor both for better relapse-free (P = 0.006), and overall survival (P= 0.03). Conclusions: These findings indicate that the presence of Foxp3+ tumor cells represents a new independent prognostic factor of improved outcome in HER2-overexpressing breast carcinoma, which could help to select high risk patients for additional therapies after neoadjuvant chemotherapy.

Generation of CHO Stable Cell Line Overexpressing HER2: an In Vitro Model for Breast Cancer
Background: Breast cancer is the most common female malignancy and the leading cause of cancer mortality in women worldwide. The human epidermal growth factor receptor2 (HER2) is a transmembrane tyrosine kinase receptor that is usually overexpressed in human breast cancers. Stable cell lines heterogeneously overexpressing HER2 are highly required as in vitro models for breast cancer research. T...
متن کاملFOXP3 expression in cancer cells and anthracyclines efficacy in patients with primary breast cancer treated with adjuvant chemotherapy in the phase III UNICANCER-PACS 01 trial.
BACKGROUND Predictive markers of response to chemotherapy are lacking in breast cancer patients. Forkhead Box Protein 3 (FOXP3) is an anti-oncogene whose absence in cancer cells could confer resistance to DNA damaging agent. So we made the hypothesis that FOXP3 expression predicts the response to anthracyclines in breast cancer patients and that adjuvant chemotherapy adding taxanes to anthracyc...
متن کاملIdentification of Prognostic Genes in Her2-enriched Breast Cancer by Gene Co-Expression Net-work Analysis
Introduction: HER2-enriched subtype of breast cancer has a worse prognosis than luminal subtypes. Recently, the discovery of targeted therapies in other groups of breast cancer has increased patient survival. The aim of this study was to identify genes that affect the overall survival of this group of patients based on a systems biology approach. Methods: Gene expression data and clinical infor...
متن کاملImmunogenicity of chimeric MUC1-HER2 vaccine against breast cancer in mice
Objective(s): Breast cancer is one of the most common cancers in the world and is on the increase. MUC1 and HER2 as tumor-associated antigens (TAAs) are abnormally expressed to some extent in 75–80% of breast cancers. In our present research, a novel chimeric MUC1-HER2 (HM) protein was designed and used to study whether an immune response can be generated against these TAAs. In vitro analysis ...
متن کاملInduction of T Regulatory Subsets from Naïve CD4+ T Cells after Exposure to Breast Cancer Adipose Derived Stem Cells
Background: Adipose derived stem cells (ASCs) provoke the accumulation and expansion of regulatory T cells, leading to the modulation of immune responses in tumor microenvironment. Objective: To assess the effect of tumoral ASCs on the trend of regulatory T cells differentiation. Methods: Peripheral blood naïve CD4+ T cells were co-cultured with ASCs derived from breast cancer or normal breast ...
